Preparation of multivalent derivatives of ergot alkaloids and their pharmacological studies

R. Gažák, P. Sedmera

A series of dimers of antiparkinsonic ergot alkaloids (terguride, pergolide) and of a clavine (pyroclavine) was prepared and fully spectrally characterized. The new compounds were tested for their antiparasitic and immunomodulatory activities. Some dimers were found to be active against the causative agent of malaria, , namely against its strains resistant to chinin. New compounds are free of negative effects and the toxicity threshold is 2 – 3 orders lower than the active concentration. These compound constitute new in the development of novel antimalarial drugs.

New cyclic oligomers of ergot alkaloids were synthesized. This is an entirely new class of ergot alkaloid derivatives.
